Signal Detection & Signal Management

From the single case to the safety signal — with a documented methodology instead of experience held in one person’s head.

What is signal detection?

Signal detection is the systematic search for indications that a medicinal product could be associated with an adverse event. From many single cases, literature references and database analyses, a signal emerges that is validated, prioritised, assessed and documented — through to the decision on whether the product information needs to be updated.

Why does it matter?

Signal management is the part of pharmacovigilance where experience most often takes the place of a process. As long as the most experienced person is there, it works. But the question an inspection asks is a different one: would your process pass scrutiny if this person were not available tomorrow? Robust here means independent of individuals — documented methodology, traceable decisions, regulated deputising.

Who needs it?

Every marketing authorisation holder, regardless of portfolio size. Particularly relevant for companies whose signal management has so far been informal, and for those facing an inspection or that have recently taken over a marketing authorisation.

What do the regulations require?

  • GVP Module IX — signal management, including validation, prioritisation and documentation
  • Articles 18 to 24 of Regulation (EU) No 520/2012 — signal management procedures
  • GVP Module V — transition from signal to risk management
  • Analysis of EudraVigilance and of your own safety database

How VigiPRO supports you

  • Signal detection as a described, repeatable process rather than a case-by-case decision
  • Signal validation and assessment with a traceable rationale
  • Complete documentation of decision paths, verifiable in audits and inspections
  • Benefit-risk assessment and derivation of risk minimisation measures
  • Role and deputy arrangements so that the process works independently of individuals

Why VigiPRO

We build the process so that it keeps running without us. The methodology is described, decisions are justified and filed — that is the difference between signal management you can show and signal management that only works as long as the right person is in the building.

Frequently asked questions

At what portfolio size is a formal signal process worthwhile?

The obligation does not depend on size. Even for a single product, GVP Module IX requires a described and documented process. The effort scales with the data volume; the requirement itself does not.

What does an inspection typically ask about signal detection?

How often analyses are performed, using which methodology, who made the decision and where that decision is recorded. Rejected signals must also be documented with a rationale — not only those that were followed up.

Can software take over signal detection?

It can pre-sort anomalies and speed up the analysis. Validation and the benefit-risk assessment remain an expert decision that must be justified and documented.
